Six years later, the Klauke team approached the same receptor from the pain side, in European Neuropsychopharmacology, identifier PMID:24210682. Oral caryophyllene reduced the late inflammatory phase of pain in the formalin test in a CB2 receptor-dependent manner, with no effect on the early phase; in a neuropathic pain model, it alleviated thermal hyperalgesia and mechanical allodynia without developing tolerance with chronic administration. In neither of the two studies was caryophyllene tested on humans or in inhaled form from dried flower; both works concern oral or systemic administration in mouse models of pain and inflammation, not sleep or mood. For the comparative side, this means that the advantage of caryophyllene in the Beach Crasher profile is a fact about the raw material composition, not a promise of any action in the patient.
What has been shown in studies on myrcene and why do the results not align in one direction?
Three studies on mice, with conclusions diverging in different directions. Intraperitoneal injection prolonged sleep while simultaneously presenting a slightly anxiogenic picture, inhalation of vapor acted anxiolytically in females, and a separate study described the abolition of pain response blocked by naloxone. The direction depends on the route of administration and the sex of the animal.
The Vale team studied sleep and anxiety behavior in 2002, and published the results in Phytomedicine under identifier PMID:12587690. Myrcene administered intraperitoneally (100-200 mg/kg) reduced the locomotor activity of mice in the open field test, prolonged sleep time induced by pentobarbital (by about 2.6 times at 200 mg/kg), and reduced the number of entries into the open arms of the elevated plus maze, which the authors described as a weak anxiogenic effect, not anxiolytic. An earlier work by the Rao team from 1990, published in the Journal of Pharmacy and Pharmacology, identifier PMID:1983154, concerned pain. Myrcene (10-40 mg/kg) reduced pain responses in mice in hot plate and writhing tests induced by acetic acid; the effect was abolished by naloxone and yohimbine, indicating the involvement of alpha2-adrenergic receptors and the release of endogenous opioids. The latest of the three, Wagner and colleagues, appeared in 2024 in NeuroSci under identifier PMID:39728677. With short puffs of vapor, mimicking inhalation in humans, beta-myrcene acted anxiolytically in the elevated plus maze in female mice, and in males only with a single puff. Unlike linalool, myrcene did not produce a synergistic effect with cannabidiol in either sex. Study model: rodent (mouse, vapor puffs).
In humans, myrcene alone has not been shown to alleviate insomnia at doses typical for inhalation of dried flower. The direction of the effect in rodents depends on the route of administration and sex: intraperitoneal injection in 2002 described a slightly anxiogenic effect, while inhalation in 2024 showed an anxiolytic effect in females. These two results do not negate each other; they simply show that the conclusion depends on conditions that no popular strain database provides.
How long does the effect last and does the strain influence this?
The influence is significantly smaller than the route of administration. Neither for Beach Crasher nor for Frosted Cherry Cookies has a pharmacokinetic study been published, so the course is described for the method of intake, not for the cultivar. The following ranges come from the description of absorption physiology and are the same for both strains.
The route of administration determines the course more than the variety itself. After vaporization, the substance passes from the lungs to the blood almost immediately, so the first sensations appear after a few minutes, the intensity increases for another ten to thirty minutes, and the whole effect lasts for two to four hours. After ingestion, the raw material first passes through the intestine and liver, so the first sensations are awaited from half an hour to two hours, and the episode can last six, sometimes eight hours. Hence the most common mistake with oral administration: anyone who thinks nothing is happening after thirty minutes and takes another dose will receive both doses at once.
